Monkeypox: A Histopathological and Transmission Electron Microscopy Study

The global outbreak of human monkeypox virus (hMPXV1) in 2022 highlighted the usefulness dermatological manifestations for its diagnosis. Infection by thus necessitated inclusion diagnostic repertoire dermatopathology. To assess histopathological and microscopical findings cutaneous lesions related to hMPXV infection, we analyzed skin biopsies from patients with positive MPXV DNA polymerase chain reaction presenting a typical course hMPXV1 infection. most prominent were ascribable pustular stage which epidermal necrosis areas non-viable keratinocytes “shadow cell” appearance evident; some cases, deep portion hair follicle acrosyringial epithelium affected. main cytopathic modifications included ballooning keratinocytes, followed Guarnieri bodies ground glass keratinocytes’ nuclei, together dense mixed inflammatory cell infiltrate neutrophil exocytosis. Transmission electron microscopy analysis demonstrated viral particle aggregates cytoplasm without any involvement nucleus. Interestingly, also found presence particles infected mesenchymal cells, although lesser extent than epithelial cells. Through this study, contributed expanding histological microscopic knowledge mpox virus, key step understanding current potential future trends disease, as well other Orthopoxvirus infections.
